The story revolves around two boys: Tsukune Aono, who inadvertently enrolls in a boarding school for monsters; and Hiro Hiyorimi, who inadvertently got involved into the Monster Realm while searching for his sister. They quickly meet a young woman who calls herself "Princess" and explains that she is, not only the Headmaster of the Yokai Academy, but a member of the royal family who are endowed with the power to turn a dead body into an immortal warrior that will serve and protect them; but she herself is semi-immortal (which is why she needs help from warriors). The story then follows Tsukune and Hiro as they assists Princess in killing other creatures that have been sent by her siblings to attack her in the competition for the Monster Kingdom's throne.
Princess has three other servants: Moka Akashiya, a vampire who soon develops an obsession with Tsukune's blood; an android named Flandre; and a half-Fire Yaksha who likes to call himself Hellboy. Later on in the story they encounter Princess' younger sister Sherwood, Sherwood has an android named Francisca. They also encounter a succubus Kurumu Kurono, a half-werewolf, named Riza Wildman, a child prodigy witch Yukari Sendo, another vampire named Reiri Kamura, a snow woman named Mizore Shirayuki and a human world witch, Ruby Tojo.
